Inside the cell, DNA polymerase cannot synthesize de novo (or “from scratch”): it can only add on to an existing 3’ end. So the cell uses the enzyme primase to first synthesize a short RNA primer, and DNA polymerase adds nucleotides onto the 3’ end of the RNA primer. In PCR, short DNA primers are provided as the starting point of DNA synthesis. For the sequence below, which primer pairs would amplify the highlighted region of DNA? Be careful, and pay attention to 5' and 3' ends.

5'3'

3'5'

A) 5'CCACGG3' and 5'GGGCTG3'

B) 5'CCCGAC3' and 5'GGTGCC3'

C) 5'CCACGGG3' and 5'GGTGCC3'

D) 5'CCACGGG3' and 5'GTCGGG3'

E) or none of the above
